I'm wondering, can a vacuum leak cause a misfire?

Yes, depending.

For example, 3rd gen Sx1's have a vacuum leak at cylinder #1 in the intake manifold gasket. It typically presents itself as a cylinder #1 misfire.

it would depend on where the leak is- emission or evap systems.i would rule out ignition system 1st and check timing,then test egr valve, pcv valve and hoses.if everything checks out spray brake cleaner or throttle body cleaner around intake and throttle body
