Yes, drain as much as possible and if you're reusing coolant, filter it before putting it back in. The highest point of the cooling system (car on a level surface) is the coolant surge tank, designed to allow automatic purging of air in the system. To lose as little coolant as possible, use the radiator petcock to drain the radiator and engine before disconnecting the heater hoses. what drains out heater hoses will be coolant in both hoses and heater core.
